Why Backbone exists
A small publication in Cambridge, Massachusetts, trying to describe peptide science accurately to people who don't work in a lab.
Backbone started in the autumn of 2021 as an email list with fewer than two hundred subscribers. Nadia Okonkwo had spent eight years covering life sciences for trade publications, most of it within walking distance of Kendall Square, and had watched the same conversation break down over and over. Someone would read a press release about a peptide therapeutic, go looking for context, and land in one of two places: a paywalled abstract written for specialists, or a forum thread where confident strangers described things that had never been tested in a human being.
Neither of those is coverage. The first assumes you already know what a receptor agonist is; the second assumes it doesn't matter. Backbone was built to occupy the space in between — the part where somebody explains what the molecule is, what the evidence actually shows, and where the evidence runs out.
The timing wasn't an accident. 2021 and 2022 were the insulin centennial: a hundred years since Frederick Banting and Charles Best's work at the University of Toronto turned a fatal diagnosis into a managed condition. That anniversary produced a wave of retrospectives, and reading them together made something obvious. Insulin is a peptide. So is oxytocin. So are half the signaling molecules keeping a person alive on any given afternoon. The word had been so thoroughly captured by supplement marketing that most readers had no idea it described ordinary, foundational biochemistry — the kind that has been winning Nobel Prizes since 1923.
So we publish explainers. Each one gets read by a biochemist before it goes out, each one links to the primary literature or to a registration on ClinicalTrials.gov where such a thing exists, and each one says out loud when the honest answer is that nobody has run the study yet. We are reader-supported, which means we don't have an advertiser whose product we'd rather not contradict.
The most useful sentence in science writing is also the least popular one: we don't know that yet, and here is what it would take to find out.

To make peptide science legible to non-specialists — the chemistry, the century of history behind it, and the regulatory machinery that decides what counts as a medicine. To correct misinformation where we find it, including our own, publicly and quickly.
And a line we don't cross: we never recommend, sell, or facilitate the use of anything. No protocols, no sourcing, no vendors, no affiliate links, and no instructions for putting anything into a body. Many of the compounds that come up in fitness and biohacking spaces are investigational — they are not FDA-approved, they are not dietary supplements, and the safety data that would justify use in healthy people has not been collected. Describing what a molecule does in a cell is journalism. Telling a reader to put it in their body is something else, and it isn't ours to do. That conversation belongs to you and a licensed clinician.

How we work
Every explainer is drafted from primary and review literature, checked against public records — regulatory approval documents, trial registrations on ClinicalTrials.gov, published guidance from the FDA — and reviewed for technical accuracy before publication. Where a claim is contested in the field, we say it's contested rather than picking the tidier version.
